We will develop a customizable interferometric scattering microscopy (iSCAT) platform with flexible illumination configurations, optimized for high-sensitivity imaging in dense and heterogeneous 3D cellular environments. This includes implementing adaptable optical geometries and coherence control to match the optical properties of different biological samples.

We will develop and validate temporal fluctuation analysis methods to extract molecule-specific physical quantities. This will involve integrating microrheology-based approaches and flow-field analysis to quantify viscoelastic properties, material density, diffusion mode, and dynamic flow characteristics at the nanoscale.

We will apply the developed platform to study biomphysics, in particular its dynamics, both in vitro and in living cells.
